The IMViC test
The Indole test · Used to determine the ability of an organism to cleave the amino acid typtophan into indole, ammonia, and pyruvic acid. · Procedures involves inoculation of a medium containing the amino acid tryptophan · If organisms has the enzyme tryptophanase, removal of the amino group from tryptophan forms indole · Indole is easily detected by the addition of Kovacs’reagents and the appearance of a cherry-red-coloured layer
The Methyl Red Test · Used to differentiate two major types of facultatively anaerobic enteric bacterial species · These organisms first metabolise glucose aerobically thus, exhausting all of the available oxygen by means of respiratory metabolism · The type of fermentations and the end products formed are of value in species identification
Voges-Proskauer · Used to detect acetoin · Is an intermediate compound produced by organisms carrying out butylene glycol type of glucose fermentation · Positive reaction àformation of pink or cherry-red colour upon the addition of alpha naphthol and potassium hydroxide creatine solutions
Citrate test · Determines the presence of enzyme that enable citrate to enter the cells and be used as a sole source of carbon for metabolism and growth · Media used for citrate testàSimmons’ citrate and Koser citrate · Simmons’ citrate agar contain the pH indicator bromothymol blue, which is green under acidic conditions and dark blue when medium becomes alkaline · Organism utilising citrate produce an alkaline reactions · Positive reaction in Koser citrate medium is indicated by the preparation becoming cloudy after incubation · A light inoculum must be used with Koser citrate to avoid false positive results
